Schizosaccharomyces cryophilus OY26 WGS sequencing

Before the official species description of Schizosaccharomyces cryophilus OY26 this strain was known as Schizosaccharomyces sp. strain OY26. The sequenced stain is a type strain and has been deposited at NRRL, Agricultural Research Service Culture Collection as NRRL Y-48691, at the Centraal Bureau voor Schimmelcultures as CBS 11777 and at NBRC, NITE Biological Resource Center as NBRC 106824. This strain was isolated as a contaminant of the Schizosaccharomyces octosporus strain CBS 7191. Genomic DNA from the S. cryophilus strain was submitted to 454 Life Sciences for whole-genome sequencing. A titanium fragment library was generated from 5 mg genomic DNA and sequenced on a single 454 run using titanium chemistry. A 3K long-tag paired-end (LTPE) library was generated in parallel from the same amount of genomic DNA and sequenced on half of an FLX run. Approximately 1.3 million high-quality reads of 400 bp were generated from the titanium run and 700 000 reads were generated from the LTPE FLX run. Quality-filtered sequences from the wholegenome shotgun fragment library and LTPE library were assembled using the 454 Newbler Assembler.
