40 genotypes of Brachypodium distachyon were sourced from the USDA germplasm repository, and grown in a common greenhouse environment at the University of Illinois. We sequenced the rhizosphere microbiomes of these plants, using a Fluidigm sequencing chip to target multiple amplicons representative of the functional diversity of these microbiomes. Primers targeted the general bacterial and fungal markers including the 16S and ITS respectively. Functional groups were additionally targeted, including nitrogen fixers with the nifH gene, AM fungi with a specific 18S primer, as well as bacteria containing the alkaline phosphatase functional gene (phoD).
