Auxenochlorella protothecoides Transcriptome or Gene expression

These data contain transcriptome profiles of the alga C. protothecoides under two different growth regimes and at three different stages of cell growth. Cultivation of the organism and total RNA extraction was conducted at the USDA-ARS Daniel K. Inouye U.S. Pacific Basin Agricultural Research Center in Hilo, Hawaii during September 2013; subsequently, samples were submitted to the Yale Center for Genomic Analysis from Illumina Hi-Seq library preparation and sequencing. Synchronized cultures were grown in either photoautotrophic medium (Bristol's salts plus 0.1% (w/v) proteose peptone) or papaya-based heterotrophic medium (P1, Bristol's salts plus 12.5% (v/v) Rainbow papaya juice). Samples containing 1.0E+08 cells were harvested from each of three biological replicate cultures grown in both medium compositions during the logarithmic growth phase. Additionally, samples were harvested from the heterotrophic cultures once the cell density had nearly saturated (the transitional point between growth and oil biosynthesis), and again during peak oil biosynthesis.
